鲨鱼软骨粉合剂诱导的抗肝血吸虫性纤维化效应的研究

摘    要:鲨鱼软骨粉(SCP)伍用“425”制剂(当归根提取物),使成鲨鱼软骨粉合剂,用以研究感染日本血吸虫3wk的BALB/c小鼠经口服用SCP合剂后4wk(相当于感染后7wk)诱导的抗血吸虫性纤维化效应及其机制。测定实验小鼠肝内血吸虫卵肉芽肿(SEG)大小和纤维化程度;用ELISA检测小鼠血清抗体IgG及其亚类IgG1水平(OD均值);应用流式细胞仪测定小鼠脾脏T细胞亚群的细胞均数和比值。实验结果:服用SCP合剂(Ⅰ组)和单用SCP(Ⅲ组)小鼠的SEG大小均显著小于感染对照鼠(Ⅳ组),且前两者肝内血吸虫性纤维化均已消失,而后者显示严重的肝纤维化。单用“425”制剂(Ⅱ组)与Ⅰ组和Ⅲ组的血吸虫卵肉芽肿减小率之间无显著性差异,但Ⅱ组仍呈现轻度的肝血吸虫性纤维化。Ⅰ组和Ⅲ组的CD4+/CD8+比值(1.70~1.88)明显高于Ⅱ组(0.95);Ⅰ组和Ⅱ组的IgG和IgG1水平均显著高于Ⅲ组和Ⅳ组。结果表明,服用SCP合剂和单用SCP小鼠,均能诱导明显的抗肝血吸虫性纤维化效应,若单用“425”制剂,则仅显示轻度的抗肝纤维化作用,其机制与细胞免疫密切相关。

Abstract:To investigate the effects and mechanism of antiS. japonicum hepatica fibrosis induced by shark cartilage powder(SCP) conbining reagent(combination of SCP with "425" reagent, namely angelica root extract), we observed hepatic fibrosis and the mean sizes of schistosome egg granuloma(SEG). At 3 weeks after infection, the infected BALB/c mice were divided into 4 groups and orally administered with SCP conbining reagent for group , "425" reagent for group and SCP for Group respectively. The group was used as a control. At 4 weeks after treatment(7 weeks post infection) the mean sizes of SEG in group and group were significantly decreased than that in group and the hepatic schistosome fibrosis in the former 2 groups disapperared already. The reduction rates of SEG had no significant difference between group and group . The ratios of CD4 /CD8 in group and group (1.70 and 1.88) were obviously higher than that in group (0.95). The IgG and IgG1 of group and group were both markedly higher than that in group and group . Therefore, shark cartilage powder could obviously induce antihepatic fibrosis effects and its mechanism was associated with cellular immune response. However, "425"reagent could induce humoral immune reponse, and little effect of antihepatic schistosome fibrosis.
