和厚朴酚联合吉西他滨协同抑制 Burkitt 淋巴瘤细胞增殖和促凋亡作用

摘    要:本研究旨在探讨和厚朴酚(Honokiol,HNK)与吉西他滨(Gemcitabine,GEM)联合应用对Burkitt淋巴瘤细胞株Raji细胞增殖和凋亡的影响.应用CCK-8法检测HNK和GEM单用及联合应用对Raji细胞增殖的抑制作用;采用流式细胞术检测两种药物诱导Raji细胞凋亡情况及细胞周期的变化;以Western blot法检测BCL-2抗凋亡蛋白水平的变化.结果表明,HNK与GEM联合后,细胞生长抑制明显高于HNK和GEM单药组(P<0.01),具有时间和浓度依赖性;细胞凋亡比例均较各单药组增多,差异具有统计学意义(P<0.01),两药联合后大部分细胞被阻滞在G0/G1期,S期细胞减少;细胞内抗凋亡蛋白BCL-2水平在联合组比在各单药组表达量下降,差异具有统计学意义(P<0.01).结论:HNK联合GEM具有协同抑制Burkitt淋巴瘤Raji细胞增殖并诱导其凋亡的作用,协同机制可能与两药联合后更多细胞发生G0/G1期阻滞,并抑制抗凋亡蛋白BCL-2的活化有关.

Abstract:This study was aimed to investigate the effect of Honokiol(HNK) combined with Gemcitabine(GEM) on the proliferation and apoptosis of human Burkitt lymphoma Raji cells.Cell proliferation was detected by CCK-8 method to study the role of Honokiol and Gemcitabine in Raji cells.The cell apoptosis and cell cycle status were analyzed by flow cytometry.The level of apoptosis-related protein BCL-2 was measured with Western blot.The results showed that compared with cells treated with mentioned above drugs alone,the proliferative potential of cells in combination group was significantly inhibited(P <0.01) and the inhibition rate was related to the concentration and action time of HNK; and apoptosis rate markedly increased(P < 0.01),while most Raji cells were arrested at G0/G1 phase and decreased in S phase after treatment with combination of two drugs; the expression of BCL-2 protein decreased (P < 0.01).It is concluded that Honokiol combined Gemcitabine can synergistically inhibit the proliferation,induce cell apoptosis,and down-regulate the expression of BCL-2 in Raji cells.The possible mechanism of synergistic effect may be related with arrest of cell cycle at G0/G1 phase and downregulation of the expression of BCL-2.
