摘    要:目的了解月见草油以不同剂量水平较长期喂养后对受试动物的毒副作用。方法选择清洁级70 g左右的SD大鼠雌雄各40只,随机分为4组,即对照组及三个受试物组,每组雌雄各10只。剂量组分别以1.034、2.069、3.103g/kg.bw剂量的月见草油灌胃90 d,对照组给予等体积植物油。于实验中期和实验末采血测定血液学指标和血生化指标。实验期间观察动物的生长发育情况、不良反应,记录摄食量,计算食物利用率。实验末取肝、肾、脾、睾丸称重后,计算脏器/体重比值;取肝、肾、脾、胃、肠、睾丸、卵巢进行组织病理学检查。结果实验期内大鼠未见明显不良反应。各剂量组大鼠体重增长、进食量、食物利用率、血常规指标、血生化指标、脏器绝对重量、脏器/体重比值与对照组比较,差异均无统计学意义(P〉0.05)。大体解剖和组织病理学检查未见与受试样品有关的异常改变。结论在本实验条件下,未见月见草油对受试动物产生明显毒副作用,90 d喂养最大未观察到有害作用剂量为3.103 g/kg.bw。

Observation on Effect of 90- day Feeding of Evening Primrose Oil on Rats

Abstract:Objective To explore the toxicity of subchronic feeding of evening primrose oil in rats.Methods Forty male and 40 female SD rats of clean grade,weighted about 70 g,were randomly divided into four groups,one control and three doses groups(each n=20).The rats in the doses groups were administered orally and respectively with evening primrose oil at 1.034 g/kg·bw,2.069 g/kg·bw,and 3.103 g/kg·bw doses for 90 consecutive days,while the control rats were administered plant oil.The general condition,body weight,food consumption and food utilization rate of the rats were monitored.Blood was collected for hematology and serum biochemistry tests in the middle of and at the end of the test.Liver,kidney,spleen and testis were collected and weighed,and then visceral organs weight/body weight ratio was calculated.Liver,kidney,spleen,stomach,intestine,testis and ovary were subjected to histopathological examination.Results No adverse reaction was found during the experiment.The weight gain,food consumption and utilization rate,heamological indicators,biochemical items and the absolute,relative weight of the main visceral organs of the rats in the doses groups did not reveal any significant differences as opposed to those of the controls(P>0.05).Gross examination and histopathological examination revealed no abnormal changes associated with the test substance.Conclusions In this test condition,administration of evening primrose oil for 90 days did not produce any adverse effects on rats.The No Observed Adverse Effect Level(NOAEL) of evening primrose oil in the 90-day feeding study was 3.103 g/kg·bw.
