多发性骨髓瘤患者骨髓基质细胞IL-1β.IL-6.SCF.TPO表达的研究

摘    要: 【摘要】 目的 研究多发性骨髓瘤 (MM) 患者骨髓基质细胞多种细胞因子在mRNA水平的表达情况,探讨不同因子在MM发生、发展中可能存在的作用。方法 采用半定量RT-PCR方法在mRNA水平上检测MM患者骨髓基质细胞白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、干细胞因子(SCF)、血小板生成素(TPO)的表达。结果 IL-1β、IL-6的表达:MM组与正常对照组和非MM组相比差异均具有统计学意义(P<0.01)。SCF的表达:自体移植后患者组与正常对照组相比差异无统计学意义(P>0.05),初发MM组和难治复发MM组与正常对照组和非MM组相比差异均有统计学意义(P<0.05)。TPO的表达:MM组与正常对照组相比差异具有统计学意义(其中初发组P<0.01,其余两组P<0.05)。初发组与非MM组相比差异有统计学意义(P<0.01),难治复发MM组和移植后患者组与非MM组相比差异无统计学意义。结论 体外培养的MM患者骨髓基质细胞 IL-1β、IL-6、SCF和TPO存在表达异常,这些因子在疾病进展中可能起重要作用。

Abstract:【Abstract】 Objective To study multiple cytokine mRNA expressions of bone marrow stromal cells in patients with multiple myeloma (MM), as well as to explore the role of these cytokines in the occurrence and development of MM. Methods Semi-quantitiative RT-PCR was used to detect the mRNA expressions of IL-1β, IL-6, SCF, TPO. Results The mRNA expression levels of IL-1β and IL-6 were higher than that of nor-mal controls and other hematological malignancies (P <0.01). For the expression of SCF, except the trans-planted MM patients, there was a significant difference when other MM patients contrast to the normal controls and other hematological malignancies (P <0.05). For the expression of TPO, there was a significant difference between the MM patients and the normal controls and between the primary MM and other hematological malig-nancies(P <0.01). There was not a significant difference when the relapsed MM and the auto-transplanted MM contrasted to other hematological malignancies. Conclusion IL-1β, IL-6, SCF and TPO on stromal cells are up-regulated at mRNA level when cultured in vitro and may play an important role in the disease pathogenesis.
