牛磺酸及复合微量营养素对大鼠视网膜iNOS mRNA的影响

摘    要:目的 探讨牛磺酸及复合微量营养素对光照和暗适应条件下大鼠视网膜iNOSmRNA的影响。方法 在幼年大鼠的饲料配方中分别添加牛磺酸及VA、VB1、VB2、尼克酸,Zn、Se等复合微量营养素,营养干预4周后,采用原位分子杂交技术检测各组大鼠视网膜iNOSmRNA在光照和暗适应条件下的分布和表达。结果 光照时iNOSmRNA分布于内,外核层及节细胞层,而暗适应时分布于内,外网状层,牛磺酸组暗适应时亦分布于内,外核层及节细胞层,微量营养素组光照和暗适应时iNOSmRNA的分布分别与普通饲料组暗适应和光照时的分布一致。结论 iNSOmRNA的分布因光适应状态而异,上述复合微量营养素能影响光照和暗适应时iNOSmRNA的分布和表达,牛磺酸则主要影响其暗适应时的分布和表达。

Abstract:Objective To explore the effects of taurine and micronutrient complex on the expression of inducible NOS(iNOS) mRNA in rat retina of rat under the condition of light and dark adaptation. Methods After supplementation of taurine, vitamin A, vitamin B 1, vitamin B 2, niacin, Zinc and Selenium in rat diet for 4 weeks, the localization and expression of iNOS mRNA in rat retina under the condition of light and dark adaptation were observed with in situ hybridization. Results iNOS mRNA was expressed in outer nuclear layer(ONL), inner nuclear layer(INL), and ganglion cells layer(GL) under the condition of light adaptation, and in outer plexiform layer(OPL) and inner plexiform layer(IPL) under the condition of dark adaptation. The changes of iNOS mRNA expression were coincident with normal group in light adaptation after taurine treatment, but the intervention of micronutrient complex induced an opposite alternation of iNOS mRNA expression to normal group in light and dark adaptation. Conclusion The localization and expression of iNOS mRNA are quite different under various condition of light adaptation. The supplementation of micronutrient complex could modulate the localization and expression of iNOS mRNA in dark and light adaptation, but taurine only modulate the localization and expression of it in dark adaptation.
