大鼠胃肠嗜银细胞的表皮角蛋白免疫细胞化学定位

本文报道应用表皮角蛋白(EK)抗体,对大鼠胃肠粘膜上皮进行了免疫细胞化学定位。应用PAP和间接免疫荧光染色技术,观察到除柱状细胞、杯状细胞呈EK阳性外,还有一种EK强阳性且在上皮内的位置以及形态酷似嗜银细胞的上皮细胞。应用浸银-PAP联合染色法显示,嗜银细胞内确实存在着EK阳性物质,位于嗜银细胞的外周部位和嗜银颗粒之间。提示胃肠嗜银细胞可能与其他胃肠粘膜上皮细胞一样,含有细胞角蛋白型中间丝(IF)。本研究的结果支持胃肠嗜银细胞起源于内胚层的假说。

IMMUNOCYTOCHEMICAL LOCALIZATION OF EPIDERMAL KERATIN IN THE ARGYROPHIL CELLS OF THE RAT GUT

In order to investigate the origin of the argyrophil cells of the gut, we made an immunocytochemical study on the gastrointestinal epithelia of rats using an antibody to bovine epidermal keratin (EK). With both PAP and immunofluorescence staining methods, we found a new kind of EK-positive epithelial cells, which were stained more strongly than the columnar or goblet cells and were very similar to the gut argyrophil cells in their shape and location. With the silver-PAP staining method which was established in our laboratory, we demonstrated that the argyrophil cells are EK-positive and the brown immunoreactive product is in the peripheral region of the cytoplasm or between the black argyrophilic granules. The results suggest that the gut argyrophil cells may contain the intermediate filaments of eytokeratin type, same as those of the columnar and goblet cells, and provide a strong support for the theory of the endoderm origin of the gut argyrophil cells.
