革皮氏海参皂苷对环磷酰胺所致骨髓损伤模型小鼠造血作用的研究

摘    要:目的研究革皮氏海参(Pearsonothuria graeffeii)皂苷对环磷酰胺所致骨髓损伤模型小鼠造血的调节作用。方法用不同剂量的革皮氏海参皂苷分别灌胃环磷酰胺所致骨髓损伤模型小鼠,采用实验血液学技术和造血祖细胞体外培养方法,检测小鼠外周血白细胞(WBC)、红细胞(RBC)、血小板(PLT)、血红蛋白(Hb)和网织红细胞(RET)数,骨髓有核细胞(BMNC),骨髓造血干细胞,粒单系祖细胞(CFU-GM),红系祖细胞(CFU-E)和巨核系祖细胞(CFU-Meg)数。结果革皮氏海参皂苷能提高小鼠外周血像中WBC、RBC、PLT、Hb和RET的数量,提高骨髓有核细胞数,促进造血干细胞的增殖和分化,增加骨髓中CFU-GM、CFU-E/CFU-E和CFU-Meg的集落生成。结论革皮氏海参皂苷对骨髓损伤模型小鼠有促进造血的作用。其作用途径可能通过调节机体的免疫功能,诱导机体产生多种造血细胞因子,促进造血干细胞的增殖,并诱导向粒单系、红系和巨核系祖细胞分化,恢复小鼠的造血功能。

Abstract:Objective To study the effect of saponin of Pearsonothuria graeffei on hematopoiesis in the myelosuppression mice induced by cyclophosphamide(Cy).Method Different dosages of saponin of P.graeffei were given to model mice of hematopoietic system impairment induced by Cy.The number of peripheral white blood cell(WBC),red blood cell(RBC),platelets(PLT),hemoglobin(Hb),reticulocyte(RET),bone marrow nucleated cells(BMNC),hematopoietic colony forming units in spleen(CFU-S),granulocyte-monocyte colony forming units(CFU-GM),colony-forming units erythroid(CFU-E),colony forming unit-megakaryocyte(CFU-Meg) were determined.Results The saponin of P.graeffei could enhance the number of peripheral blood WBC,RBC,PLT,Hb,RET and BMNC,promote the proliferation and differentiation of hemopoietic stem cells(HSC),increase the number of CFU-GM,CFU-E/CFU-E,and CFU-Meg in the myelosuppression mice.Conclusion The saponin of P.graeffei has the function of hematopoiesis in the myelosuppression mice induced by Cy.Its approaches may be related to regulating immune function,inducing production of many hematopoietic cellular factors,reducing time of cell cycle to promote the proliferation and differentiation of HSC,CFU-GM,CFU-E/CFU-E and CFU-Meg and recover the function of hematopoiesis in mice.
