硒和锶明胶纳米微粒的制备及其代谢动力学

摘    要:目的 : 制备微量元素硒 (Se)和锶 (Sr)明胶纳米微粒 ,研究它们的体内代谢动力学。方法 : 用复凝聚法制备 Se和 Sr明胶纳米微粒 ,用透射电子显微镜负染法观察明胶纳米微粒的大小、形态 ;用原子吸收分光光度法测定 Se和 Sr明胶纳米微粒中 Se和 Sr的包裹率及犬血液中的含量 ;用 3 P87药代动力学程序计算 Se和 Sr代谢动力学参数。结果 : 用复凝聚法制备的 Se和 Sr明胶纳米微粒粒径为 (1 45± 1 5 ) nm和 (1 5 6± 2 2 ) nm,Se和 Sr的包裹率分别为 85 .4%和 82 .7% ,与单纯 Se和 Sr对照组相比 ,Se和 Sr明胶纳米微粒的体内滞留时间延长 2~ 4倍 ,生物利用度提高1 .6~ 2 .2倍。结论 : Se和 Sr制成明胶纳米微粒后其生物活性好 ,具有缓释作用 ,提高了它们在体内的滞留时间和生物利用度。

Abstract:Objective:To prepare the gelatin selenium and strontium nanoparticles(TENP) and investigate their pharmacokinetics . Methods: The entrapment efficiency of gelatin Se and Sr nanoparticles and their content in dog plasma were measured by atomic absorption spectroscopy method. The pharmacokinetic parameters were calculated by 3P87 computer program. Results: The diameter of gelation Se and Sr nanoparticles prepared were (145±15) nm and (156±22) nm respectively. The entrapment efficiency of Se and Sr of gelatin nanoparticles were 85.4%and 82.7%. Compared with the free Se and Sr group(control group),the release time of Se and Sr gelatin nanoparticle in vivo was 2 and 4 times longer respectively, and their bioavailability was increased 1.6 and 2.2 times. Conclusion: The selenium and strontium gelatin nanoparticles can preserve the biological activies of trace elements and enhance their time and bioavailability in vivo.
