摘    要:目的 探讨蝎毒对放疗的增敏效应及其作用机制。方法 将Lewis肺癌细胞接种于NIH小鼠右后肢外侧皮内,当肿瘤平均直径达到6mm左右时开始实验,观察蝎毒(0.6mg/kg)、放疗(4Gy)及其联合治疗组的肿瘤倍增时间(MDDT)、生存时间(AST)、病理改变和细胞凋亡变化。结果 对照组、蝎毒组、放疗组和蝎毒→放疗组的MDDT分别为(10.2±0.34)d、(11.5±0.38)d、(13.7±0.42)d和(16.1±0.53)d,各治疗组均可延长荷瘤小鼠生存期,引起肿瘤组织的坏死,诱发细胞凋亡,联合治疗效果更为突出。结论 蝎毒对荷Lewis肺癌的NIH小鼠瘤体不仅有抑制或杀伤作用,而且对放疗有增敏作用。诱发细胞凋亡是其抗肿瘤的主要机制之一。

Abstract:Objective To determine the radiosensitization effect of scorpion toxin(ST)and its mechanism.Methods The Lewis lung car cinoma cells were injected subcutaneously in right hind leg of the NIH mice.Treatments were began when the tumors reached about 6mm in diameter.The effects of ST(0.6mg/kg),radiation(4Gy,RT)and their combination group(ST→RT)on mean diameter doubling time (MDDT),animal survival time(AST),tumor necrosis and apoptosis were observed.Results The MDDT of the control group and ST,RT and ST→RT groups were(10.2±0.34)days,(11.5±0.38)days,(13.7±0.42)days and (16.1±0.53)days,respectively.In all treatment groups AST of the mice bearing Lewis lung carcinoma was prolonged, tumor necrosis and apoptosis were induced. The combination of radiation and drug had a strong synergetic effect.Conclusion Scorpion toxin(0.6mg/kg)has both antitumor effect and sensitizing effect to radiation(4Gy).Induced apoptosis is one of the antitumor mechanisms.
