摘    要:目的 观察三苯氧胺(amoxifen,TAM)及雌激素(E2)对乳腺癌MCF-7细胞株耐药基因mdr-1mRNA水平的影响:方法 应用RT-PCR法观察TAM及E2在不同时间、浓度作用下细胞内mdr-1mRNA水平的变化;结果TAM可以使细胞内mdr-1mRNA水平降低,而E2则相反。且以上变化表现出剂量、时间依赖关系。结论 TAM通过降低mdr-1mRNA水平,使肿瘤细胞的耐药性降低,有利于抗肿

Abstract:Objective Study the effects of Tamoxifen(TAM) and estrogen(E2) on the mdr-1 mRNA level in MCF -7 cell. Methods RT-PCR was used to observe the variation of mdr-1 mANA level in MCF-7 cell after treatment with TAM and E2 at different time and concentration. Results TAM could decrease the level of mdr-1 mRNA in MCF-7 cell but E2 could not. These results came together with the relationship with the concentration of the drug and the treatment of the given time. Conclusion The drug-resistance of neoplasm cells could be down -regulared by TAM via decreasing the mRNA level in cells, which would be helpful in tumor drug therapy.
