辛基酚对MCF-7乳腺癌细胞增殖及凋亡作用研究

摘    要:目的探讨辛基酚(octylphenols,OP)诱导乳腺癌细胞凋亡及抑制乳腺癌细胞增殖的可能机制。方法应用MTT法及流式细胞仪检测OP对乳腺癌细胞MCF-7的增殖抑制及细胞周期的变化;Real Time PCR和Western blot法检测OP对MCF-7乳腺癌细胞中细胞周期相关蛋白p27Kip1和CyclinE的mRNA及蛋白表达的影响及对凋亡抑制蛋白PAK4表达的影响。结果 OP对MCF-7细胞增殖有明显的抑制作用,MCF-7细胞阻滞在G0/G1期,使S期的细胞减少;Real Time PCR和Western Blot检测发现培养的MCF-7细胞,随着OP的处理浓度增加,CyclinE蛋白在分子水平和蛋白水平均降低,p27Kip1的表达量升高。结论 OP可能通过调控细胞周期相关蛋白的表达,抑制MCF-7乳腺癌细胞增殖。

Abstract:OBJECTIVE To investigate the possible mechanism by which octylphenols (OP) induces apoptosis and inhibits proliferation of human breast cancer cells. METHODS MTT assays and flow cytometry analysis were employed to examine cell proliferation and cell cycle distribution, respectively. The expression levels of cell cycle associated protein p27Kip1 and CyclinE were determined by Real time PCR and Western blot analysis. The protein level of PAK4, an apoptosis inhibitor, was also analyzed. RESULTS OP significantly inhibited the growth of MCF-7 cells and caused G0/G1 cell cycle arrest in MCF-7 cells. In addition, after treated with OP, the percent of cells in S phrase was reduced. OP treatment decreased the expression of CyclinE at both mRNA and protein level in a dose-dependent fashion. However, elevated expression of p27Kip1 was observed after OP treatment. CONCLUSION OP may inhibit the proliferation of MCF-7 cells by regulating the expression cell cycle associated proteins.
