膀胱癌中IGF-2和H19基因特异表达模式及印记缺失分析

摘    要:目的:探讨胰岛素样生长因子-2(IGF-2)和H19等位基因在膀胱癌中特异性表达的模式及基因组印记丢失的分析。方法:结合PCR和限制性片段长度多态性分析RFLP技术以及DNA甲基化检测技术分析我院收集的膀胱癌标本IGF-2和H19等位基因进行杂合子筛选并进一步研究基因印记缺失与膀胱癌两者之间的关系。结果:在40例膀胱癌中IGF-2等位基因筛选出21例杂合子现象的标本,在这21例杂合子标本中有12例发生了基因组印记缺失;H19等位基因筛选出19例杂合子现象的标本,在这19例杂合子标本中有10例发生了基因组印记缺失。H19基因缺失现象与H19DMR区域的高度去甲基化正相关,而IGF-2基因缺失现象与H19DMR区域的高度甲基化正相关。结论:膀胱癌中IGF-2和H19的基因印记缺失与H19DMR区域的甲基化状态有高度的相关性。

ANALYSIS OF THE SPECIFIC-EXPRESSING AND LOSS OF IMPRINTMG OF IGF-2 AND H19 GENE IN BLADDER CANCER

Abstract:Objective:To investigate status of specific-expressing and loss of imprinting of insulin like growth factor-2(IGF-2) and H19 allele gene.Methods: With PCR-RFLP and bisulfite treatment of genomic DNA,the loss of imprinting of IGF-2 and H19 gene specific-expressing and the filter of heterozygosity of chromosome in 40 bladder cancer samples in our hospital were analyxed.Results: It was identified that LOI of IGF-2 gene in 12 bladder cancer samples and LOI of H19 gene in 10 bladder cancer samples among 40 bladder cancer samples.LOI of H19 gene could be linked to hypomethylation of H19 DMR.and that LOI of IGF-2 gene could be linked to hypermethylation of H19 DMR.Conclusion:The loss of imprinting of IGF-2 and H19 gene could be linked to the methylation state of H19 DMR in bladder cancer.
