中药葛根对去卵巢大鼠骨代谢生化指标的影响

摘    要:目的 研究中药葛根对去卵巢大鼠骨代谢生化指标的影响 ,探讨葛根治疗骨质疏松症的药理作用。方法 采用去卵巢大鼠模型 ,分A去势组、B假手术组、C去势服葛根组、D去势服西药组 (阳性对照组 ,程序治疗 ) ,去势 1个月后C、D组服药 50d ,放免法测定血清雌二醇 (E2 )、邻甲酚酞络合酮法测定尿钙 ,ELISA法测定尿脱氧吡啶啉 (Dpd) ,放免法测定血清骨钙素 (BGP)水平 ,组间进行比较。结果 E2 测定B、C、D组高于A组 (P <0 0 0 1 ) ,C、D组低于B组 (P <0 0 1 ,P <0 0 0 1 )。尿钙测定B、C、D组低于A组 (P <0 0 1 )。Dpd测定B、C、D组低于A组 (P <0 0 0 1 ) ,C、D组低于B组 (P <0 0 5 ,P <0 0 1 )。BGP测定B组低于A组 (P <0 0 1 ) ,C、D组高于A组 (P <0 0 1 ,P <0 0 5) ,同时高于B组(P <0 0 0 1 )。结论 中药葛根有增加E2 ,抑制骨吸收 ,促进骨形成 ,对骨质疏松症有治疗作用

Abstract:Objective To investigate the therapeutic mecha ni sm of Pueraria lobata (Willd ) Ohwi in osteoporosis, and its effects on bon e metabolism in ovariectomized rat Methods The female rats we r e divided into four groups A was ovariectomized group, B sham operation group, C Pucraria group, D programmed treatment group, or positive control group One m onth after ovariectomy, groups C, D were treated with relevant drugs for 50 days , serum estrogen (E 2) and osteocalcin (BGP) levels were determined by RIA, uri ne calcium level was determined by orthocresolphalein complexon (OCPC) method, u rine deoxypyridinoline (Dpd) was measured by ELISA Results E 2 levels in groups B, C, D were higher than that in group A (P<0 001), and those in group C, D were lower than in group B (P<0 01,P<0 001) Urine calcium levelsin groups C and D were lower than that in group A (P <0 01) D pd levels in groups B, C, D was lower than that in group A (P<0 001), thes e in groups C, D were lower than that in groups B (P<0 05,P<0 01) BGP level in group B was lower than that in group A (P<0 01), those in groups C , D were higher than that in grpus A (P<0 01,P<0 05), and higher tha n those in groups B (P<0 001) Conclusion Pueraria loba ta (Willd) Ohwi increases serum E 2 level, inhibits bone resorption, induces bo ne formation, so it has therapeutic effect on osteoporosis
