导流杂交基因芯片技术在肠道致病菌检测中的应用研究

摘    要:目的建立导流杂交芯片技术检测粪便标本中8种肠道致病菌。方法以细菌16S rDNA和23S rDNA序列设计通用引物和寡核苷酸探针,将活性氨基标记的探针依次固定在BiodyneC膜上,制成检测芯片,用标记生物素的引物进行PCR扩增,将扩增产物与检测芯片在核酸分子快速杂交仪上进行杂交,以POD和TMB进行显色,判断结果;应用该方法检测8种肠道致病菌标准菌株和120份腹泻患者粪便标本,通过细菌培养结果比较该方法的有效性。结果120份临床粪便标本中,采用导流杂交芯片共检出68份(56.67%)携带肠道致病菌;细菌培养鉴定出63份(52.50%)携带肠道致病菌,其中58份导流杂交芯片与细菌培养结果一致,准确率为85.29%。结论导流杂交芯片技术能够快速准确检测8种常见肠道致病菌,而且适用于流行病学调查研究。
