低分子肝素明胶纳米微球的制备及条件比较

摘    要:目的研制低分子肝素明胶纳米微球,并对其制备条件进行比较性研究。方法用乳化凝聚法制备低分子肝素明胶纳米粒,并用正交设计优化制备工艺,用电子扫描显微镜观察纳米粒表面形态,紫外分光光度法测定纳米粒的包封率。结果低分子肝素明胶纳米粒为圆形或椭圆形,分散性好,粒径40~100 nm,包封率在80%以上,最佳制备工艺重现性良好。结论以明胶为囊材,采用乳化凝聚法,制备了低分子肝素明胶纳米粒,且制备工艺简便稳定,具有应用前景。

Abstract:Purpose To prepare low molecular weight heparin gelatin microsphere,and to do comparative research about the preparation condition.Methods Low molecular weight heparin gelatin nanoparticles were prepared through the way of emulsification and agglomeration ,the optimal technique were chosen by orthogonal design,their superficial shape was observed under the electronic scanning microscope,and the encapsulated ratio of nanoparticles was determined by ultraviolet spectrophotometry.Results Gelatin nanoparticles of low molecular weight heparin were globular or elliptic,and with well dispersibility,the diameter scope was between 40 and 100nm,and the encapsulated ratio was more than 80 percent,and the optimal technique has a good reproducibility.Conclusion The technique of preparing low molecular weight heparin gelatin nanoparticles by the method of emulsification and agglomeration with the material of gelatin is convenient,and stable.It has an applied perspective.
