摘    要:雄性鹌鹑用高脂饲料喂养8wk能形成典型的高脂血症和动脉粥样硬化斑块,给动物po硫酸苷糖酯(PGMS)50及25 mg·kg~1·d~1,能明显抑制鹌鹑血清总胆固醇TC及(LDL+VLDL)C水平的升高,提高HDL C/TC及HDL_2/HDL_3 C比值,实验8 wk末,高脂组动脉病变重,斑块发生率为91.7%(11/12);PGMS高,低剂量组动脉病变轻,斑块发生率分别为25.0%(3/12)及41.7%(5/12).光镜和电镜观察表明,用药组动脉内膜增生明显轻于高脂组,提示硫酸甘糖酯对平滑肌细胞的增生具有抑制作用,并能延缓动脉粥样硬化的发生和发展。

Abstract:Propylene glycol mannate sulfate(PGMS)is a new type of low molecular weight heparinoid marine agent first developed in China.In this paper,the preventive effects of PGMS on exprimental atherosclerosis were investigated in quails.Healthy male Korean quails were randomized into 4 groups: group I was fed with stock diet; group Ⅱ,hypercholesterol diet; group Ⅲ,hypercholesterol diet plus PGMS 50mg·kg-1·d-1 po and group Ⅳ,hypercholesterol diet plus PGMS 25 mg·kg-1·d-1 po.Total serum cholesterol(TC),HDL-C and HDL3-C were determined by the enzymatic method at the end of 6 wk.HDL2-C,(LDL+VLDL)-C,and the ratios of HDL-C/TC and HDL2-C/HDL3-C were calculated.The aorta and the left and right brachiocephalic arteriae were dissected out by the end of wk 8.The samples were stained with Sudan Ⅳ and the plaque incidence was calculated before the examination under the light and electron microscopes.Results show that PGMS significantly inhibits the elevation of serum TC and(LDL+VLDL)-C,and increases the HDL-C/TC and HDL2-C/HDL3-C ratios.The atherogenic lesions in hypercholesterol group were severe with the incidence of atherogenic plaque being 91.7%(11/12),and mild in both PGMS-treated groups with the incidence of 25.0%(3/12)and 41.7%(5/12),respectively,and therefore the lesions in both treated groups were significantly different from those in the hypercholeslerol group.Histological examinations showed that there were 5-10 layers of foamy cells in the hypercholesterol group and only 2-5 layers of foamy cells in both PGMS-treated groups.Electron microscopic examinations showed that lipid droplets inside the smooth muscle cells in PGMS-treated groups were fewer thanthose in hypercholesterol group,suggesting that PGMS can inhibit the proliferation of smooth muscle cells and prevent the development of exprimental atherosclerosis.
