摘    要:目的:建立乙型肝炎病毒(HBV)基因型的新分型方法。方法:对HBV全基因序列和S基因片段序列,分别建立系统进化树作基因分型;同时,利用S基因片段序列中基因型特异性核苷酸,建立一种较为简单的基因分型方法。结果:S基因区进化树与全基因进化树在基因族的分布上基本相似。在S基因区,基因型A在核苷酸(nt)451位有型特异核苷酸T;基因型B在nt321位有A,nt324位有T,nt345位有G,nt408位有G;基因型C在nt293位有A,nt294位有C,nt491位有A;基因型D在nt290位有A;基因型E在nt619位有A;基因型F在nt287位有C,nt288位有T,nt294位有G,nt337位有T;基因型G在nt290位有G,nt306位有T,nt347位有T,nt481位有G。对18例基因型B和基因型C分子进行树进行分析,所得的基因型结果与S基因型特异核苷酸方法相同。结论:我们建立的一种依赖基因型特异性核苷酸的HBV基因分型方法,经分子进化树分析证明方法可靠。

Hepatitis B virus genotype assignment based-on genotype specific nucleotide in S region
Abstract:Objective To establish a simple genotyping method. Methods Phylogenetic trees of full-length HBV genome and S gene were used to verify genotypes. The genotype-specific nucleotide in S region were searched and used for genotyping.Results The distribution of gene clusters between the phylogenetic trees of full-length HBV genome and S gene are similar, there are special nucleotides thymine (T) at nt451 in S region of genotype A; adenine (A) at nt321, (T) at nt324, guanine (G) at nt345 and nt408 of genotype B; (A) at nt293, cytosine (C) at nt294, (A) at nt491 of genotype C; (A) at nt290 of genotype D; (A) at nt619 of genotype E; (C) at nt287, (T) at nt288,(G) at nt 294, (T) at nt337 of genotype F as well as (G) at nt290, (T) at nt 306 and nt347, (G) at nt481 of genotype G. HBV genotyping with these special nucleotide is consistent with genetic analysis using the S region phylogenetic tree in 18 genotype B and C isolates. Conclusion A novel genotyping method based on genotype-specific nucleotide was developed. The reliability of the method was verified by phylogenetic tree.
