Abstract:Objective To investigate whether 5-Allyl-7-gen-difluoromethoxychrysin(AFMC) selectively enhances the cytotoxic effect of human lung cancer A549 cells to TNF-related apoptosis-inducing ligand(TRAIL).Methods Human lung cancer A549 cell line and human embryonic lung WI-38 cell line were cultured in vitro.The lactate dehydrogenase activity was evaluated by automatic biochemisitry analyzer.The apoptotic rate was examined by flow cytometry using PI fluorescence staining.Results AFMC synergetic act with TRAIL increased the cytotoxicity in human lung cancer A549 cells by assay based on a LDH release.The CI value for combination of AFMC and TRAIL was 0.019.In embryonic lung WI-38 cells by co-treatment with AFMC and TRAIL at suboptimal concentration had no apparente cytotoxicity.Flow cytometry analysis with PI staining indicated that the apoptotic rate in human lung cancer A549 cells by co-treatment with AFMC and TRAIL at suboptimal concentration for 24 h was 37.80% ± 1.78%.The apoptotic rate in human embryonic lung WI-38 cells at the same condition was 0.35% ± 0.12%.Conclusion AFMC at subtoxic concentration possess augmentation of TRAIL induced cytotoxicity in human lung cancer A549 cells.
