人血清中补体成分C_1~-抑制物抗原的分离

摘    要:补体活性第一组份抑制物(Cinhibitor,以下称C-INH)与C大分子中的C1和C1结合后,抑制了C的酶活性,并将C1和C1从C大分子中解离下来,形成C1和Cl(C—INH)_2复合物(以下称此复合物为C—INC)。C—INC具有不被强变性剂如尿素、十二烷基磺酸钠、盐酸胍等所分离的性质。据以上事实,我们用抗人Cls亲和柱分离人血清中仍具Cls抗原性的C—INC;在制备抗C—INC亲和柱的基础上,分离血清中C-INH和Cls/Clr的混合物,然后用抗Clrs亲和柱吸收混合物中的Cls/Clr,从而获得了具补体经典通路溶血抑制活性的Ci-INH样品,并制备了兔抗人C-INH抗血清。

Abstract:A new method of purifying human serum C1 inhibitor by using indirect affinity chromatography is reported in this paper. This method is based on the formation of Cl? C1s (C1-INH ) a complex of C1 and C1-INH. An anti-CIF Cls (C1-INH)2 affinity chromatography column was made after the complex was isolated from human serum by using anti-C1s affinity chromatography column. Under certain condition, the mixture of Cls, Clr and CJ-INH was isolated from human serum by anti-C1r C1 (C1-INH)2 affinity chromatography column. Then the Cls, Clr in the mixture was absorbed by anti-Clrs affinity chromatography column. In this way, C1-INH which remained its biological activity was obtained. Anti-human C1-INH serum was then prepared in rabbits
