| 摘 要： | 探讨去甲基化试剂5-氮胞苷对EBV阳性胃癌细胞系的特异性去甲基作用。方法：用逆转录-聚合酶链式反应（RT-PCR）和PCR-southern检测不同剂量5-氮胞苷处理前后EBV启动子的活性,并用Westernblot检测EBV相关基因的表达。结果：5-氮胞苷处理后EBV阳性胃癌细胞系中被甲基化失活的C启动子重新活化,并表达LMP1（潜伏性膜蛋白1）,且呈剂量依赖性。结论：5-氮胞苷可能对EBV阳性胃癌的特异性治疗有帮助。

| Abstract: | OBJECTIVE:To explore if 5-azacytidine was useful to the special treatment of EBV positive gastric cancer.METHODS:RT-PCR and PCR Southern blot were used to detect different activation of EBV EBNA promoter,and Western blot was used to check LMP1(latent membrane protein 1)expression with or without different concentrations of 5-azacytidine treatment.RESULTS:After being treated with 5-azacytidine,methylated EBV C promoter in EBV positive gastric cancer cell lines was found to be active,and the expression of LMP1 was also found to be augmented with the increasing concentrations of 5-azacytidine.CONCLUSION:5-azacytidine may be helpful to the special treatment of EBV positive gastric cancer. |
