海藻糖和葡萄糖联合负载红细胞的效果评价

摘    要:目的探讨海藻糖和葡萄糖联合负载红细胞的效果,为冷冻干燥红细胞提供新的保存剂。方法分别采用0、0.125、0.25、0.5和1mol/L的海藻糖、葡萄糖以及海藻糖联合葡萄糖37℃负载红细胞6h。分别检测负载后红细胞内海藻糖和葡萄糖的浓度。结果负载液中糖类浓度为0.125、0.25和0.5mol/L时,联合负载组与海藻糖或葡萄糖组负载后红细胞内的海藻糖和葡萄糖浓度的差异无统计学意义(P〉0.05)。而负载液糖类浓度达到1mol/L时,联合负载组红细胞内海藻糖和葡萄糖的浓度明显低于海藻糖和葡萄糖单独负载组,差异有统计学意义(P〈0.05)。结论浓度小于1mol/L时,海藻糖联合葡萄糖负载红细胞并不影响海藻糖和葡萄糖进入红细胞内,可以满足红细胞冷冻干燥的要求。

Abstract:Objective To explore new preservation agent for lyophilization of red blood cells. The effects of trehalose and glucose on loading red blood cells were studied in the present study. Methods The red blood cells were loaded with 0,0. 125,0. 25,0. 5 and 1 mol/L trehalose, glucose and combination of trehalose and glucose respectively in 37℃ for 6 hours. The levels of trehalose and glucose in the loaded red blood cells were analyzed respectively. Results There were not significant differences of trehalose and glucose levels in the loaded red blood cells among the three treatment groups (0. 125,0. 25,and 0.5 mol/L trehalose, glucose, combination of trehalose and glucose). However, the concentrations of trehalose and glucose in the red blood cells loaded with combination of trehalose and glucose (1 mol/L) significantly de- creased than those of 1 mol/L trehalose or glucose treatment alone. Conclusion It suggested that loading with combination of trehalose and glucose (〈1 tool/L) had no significant influence on the concentrations of trehalose and glucose in the loaded red blood cells. The combination of trehalose and glucose was qualified for loading red blood cells for purpose of lyphilization.
