| 摘 要： | 目的 检测乳腺定位表达载体在动物乳腺中的表达活力。方法与结果 将乳腺定位表达载体乳清酸蛋白(人组织型纤溶酶原激活剂（WAP-tPA）通过乳腺导管直接注入妊娠后期小鼠的乳腺中,待小鼠产子、泌乳后,检测出小鼠乳汁中tPA的表达量为80 ng/ml。结论 此方法可以作为从动物整体水平上检测乳腺定位表达载体表达效力的一种简便而有效的方法。

| Abstract: | Objective To assess the expression ability of WAP-tPA plasmid, which was designed to be expressed specifically in the mammary glands. Method and Results The plasmid was injected directly into the mammary glands of the mice in later gestational period through the papillary ducts to transfect the glandular cells in vivo. The milk of the mouse in lactation period was obtained and the tPA expressed in the milk was detected at a level of 80 ng/ml. Conclusion It is a simple and effective method for testing the expression ability of special vectors by transfecting the vectors directly into the mammary glandular cells in vivo. |
