摘    要:【目的】观察葛根素对大鼠颈动脉球囊损伤术后血管再狭窄形成过程中基质金属蛋白-2(MMP-2)、1型胶原(Collagen1)及细胞外信号调节激酶(ERK)表达的影响。【方法】采用球囊扩张方法,建立大鼠颈总动脉损伤模型。20只Wistar大鼠随机分为单纯球囊扩张组(对照组)和球囊扩张+葛根素治疗组(治疗组)。球囊损伤术后,高脂饲料喂养4周后观察血管形态学变化,免疫组织化学法检测MMP-2和Collagen1水平,Western blot方法检测ERK磷酸化水平。【结果】治疗组较对照组管腔面积明显增大,新生内膜面积明显减少(P〈0.05);MMP-2和Collagen1表达降低,ERK磷酸化水平降低(P〈0.01)。【结论】葛根素能抑制大鼠球囊损伤部位再狭窄,该作用可能与葛根素能降低MMPL2和Collagen1的表达及ERK磷酸化水平有关。

Abstract:Objective] To investigate the effects of puerarin on matrix metalloprotenase-2 (MMP-2), collagen type 1 (collagen 1) and extraeellular signal-regulated protein kinases (ERK) during the procedure of vascular restenosis after balloon injury. Methods] Balloon dilation was used to establish the neointimal injury model of left carotid artery in rats. Twenty Wistar rats were randomly divided into single balloon dilation group (control group) and balloon dilation followed by puerarin therapy group (treatment group). Rats were treated with drugs and high fat foods for 4 weeks. Vascular morphological changes were determined. Immuno- histochemistry method was done tO detect the expression of MMP-2 and collagen 1. Western blot assay was used to test the activation of ERK. Results] Lumem area was larger and the neointima area was smaller in the treatment group than that in control group ( P 〈0.05). The expression of MMP-2 and collagen 1 was decreased, and the activation of ERK was suppressed in the vascular in the treatment group ( P 〈0.01). Conclusion] Puerarin can inhibit vascular restenosis in rabbits by modulating the expression of MMP-2 and collagen 1 and phosphorylation of ERK.
