PTEN基因在人和树鼠句肝细胞癌组织中低表达

摘    要:目的:研究人和树鼩肝癌和癌旁组织中PTEN基因的表达情况以及与肝癌侵袭转移的关系。方法:用RT-PCR和免疫组织化学方法分别检测人肝癌、癌旁组织和正常组织以及树鼩肝癌和癌旁组织中PTEN mRNA和PTEN蛋白的表达情况。结果:PTEN mRNA在人的正常组织、癌旁组织、肝癌组织中的表达依次降低,差异有统计学意义,t值分别为-0.955和-10.065,P值均〈0.001;树鼩肝癌组织中PTEN mRNA的表达显著低于癌旁组织中的表达,t=-3.25,P=0.004。PTEN蛋白的表达在人的癌旁组织显著高于肝癌组织,t=-2.163,P=0.038;在树鼩则无差别,t=0.757,P=0.459。结论:PTEN基因的表达缺失可能与肝癌的发生有关;用树鼩进行肝癌发病学的研究基本上是可行的。

Abstract:OBJECTIVE:To study the expression of gene PTEN in human and tree shrew heptocellular carcinoma and peritumoral tissues and its relation to invasion and metastasis of liver cancer.METHODS: RT-PCR and immunohistochemistry were employed for the detection of expression of PTEN-mRNA in human heptocellular carcinoma, peritumoral tissues and normal tissues and tree shrew heptocellular carcinoma and peritumoral tissues, respectively.RESULTS:The expression of PTEN-mRNA in human heptocellular carcinoma, peritumoral tissues and normal tissues declined in turn, showing significant differences in statistics (t=0.955,P<0.001;t=-10.065,P<0.001); whereas that in tree shrew heptocellular carcinoma appeared significant lower level than that in its paratumoral tissues, t=-3.25,P=0.004. The expression of PTEN protein in human peritumoral tissues appeared the significantly higher level than that in human heptocellular carcinoma,t=-2.163,P=0.038; in contrast, no significant difference was found for tree shrew group, t=0.757,P=0.459.CONCLUSION: Deficiency in the expression of gene PTEN possibly correlates with hepatocellular carcinogenesis, and it is basically feasible to use tree shrews to study etiology of liver cancer.
