羊栖菜多糖对血管内皮细胞增殖活性的影响

摘    要:目的研究羊栖菜多糖(SFPS)对人脐静脉内皮细胞(HUVECs)增殖活性的直接作用及其对氧化损伤的内皮细胞增殖活性的影响。方法采用四甲基偶氮唑盐(Mar)比色法检测羊栖菜多糖对内皮细胞增殖活性的影响。结果SFPS在较低浓度(10mg/L~300mg/L)时,对HUVECs增殖活性具有促进作用;而当浓度过高(达到1000mg/L)时,则反而抑制细胞的增殖活性。10mg/L~100mg/L的SFPS对H2O2引起的内皮细胞增殖率下降可起到明显的保护和修复作用。结论一定浓度的羊栖菜多糖可增强血管内皮细胞增殖活性,并可对抗H2O2对此活性的抑制作用。高浓度的羊栖菜多糖抑制血管内皮细胞增殖活性。

Abstract:ObjectiveTo study the effect of sargassum fusiforme polysaccharides (SFPS) on proliferate activity of human umbilical vein endothelial cells(HUVECs) and protective and repairing effect of SFPS on HUVECs' proliferate activity injured by H_2O_2. MethodsProliferate activity of HUVECs was assessed by MTT assay. Results SFPS increased proliferate activity of HUVECs at lower concentration(10mg/L300mg/L), but reduced proliferate activity at high concentration(1000mg/L). Furthermore,at lower concentration SFPS had significant protective and repairing effect on proliferate activity of HUVECs injured by H_2O_2. Conclusion At definite concentration SFPS not only increased proliferate activity of HUVECs but also resisted injury on HUVECs by H_2O_2.
