| 摘 要： | 目的:探讨甲基转移酶DNMT1在混合性结缔组织病(MCTD)患者外周血单个核细胞(PBMC)中的表达及其意义。方法:分离MCTD患者及正常人群PBMC,采用Qiagen RNA/DNA mini kit提取RNA,实时-定量PCR检测DNMT1mRNA的表达。结果:MCTD患者PBMC中甲基转移酶DNMT1 mRNA表达较正常对照组明显降低,两组比较有统计学意义(P<0.001)。结论:MCTD患者PBMC中甲基转移酶DNMT1表达水平显著降低,DNMT1可能在MCTD发病中起作用。

| Abstract: | Objective: To investigate the significance of DNA Methyltransferase 1 mRNA expression in PBMC of mixed connective tissue disease(MCTD).Methods: 20 patients with MCTD and 10 healthy controls were recruited,PBMC were isolated by Ficoll-Hypaque density gradient centrifugation,the mRNA level of DNMT1 was measured by real-time quantitative polymerase chain reaction.Results:DNMT1 mRNA level was significantly decreased in PBMC of Mixed Connective Tissue Disease compared with healthy controls(P<0.001).Conclusions:Reduced DNA methylation and abnormal expression of methylation-related genes in PBMC may be associated with the pathogenesis of MCTD. |
