中人氟安对人肝癌裸鼠异位移植瘤生长的抑制作用

摘    要:目的:探讨中人氟安对人肝癌裸鼠异位移植瘤生长的影响。方法:建立人肝癌裸鼠异位移植瘤模型55只,随机分为阴性对照NS 0.1ml组(NS),阳性对照5-FU200mg/kg组(5-FU),中人氟安100mg/kg组(FIL)、中人氟安200mg/kg组(FIM)和中人氟安300mg/kg组(FIH),各组均为肿瘤局部给药。于治疗前及治疗后15d内多次测量肿瘤大小,绘制肿瘤生长曲线图;给药后15d处死小鼠剥离肿瘤称瘤重,计算抑瘤率;观察小鼠实验前后的生长情况及体重变化;收集血样本查血常规。结果:FIH组小鼠用药1周内全部死亡,故未纳入统计结果分析,NS组、5-FU组、FIL组、FIM组平均肿瘤体积分别为:(842.78±100.15)、(408.25±40.68)、(313.12±18.53)、(171.74±10.09)mm3,FIM组抑瘤率73.5%,与NS组抑瘤率0%比较差异有统计学意义(P<0.01),与5-FU组抑瘤率46.0%比较差异有统计学意义(P<0.05);FIL组抑瘤率52.4%,与5-FU组抑瘤率46.0%比较差异无统计学意义(P>0.05);FIM组与FIL组抑瘤率比较差异有统计学意义(P<0.05)。小鼠体重及小鼠体重增加两项指标,不同剂量实验组与NS组的比较其差异均无统计学意义(P>0.05);不同剂量实验组与5-FU组比较,差异均有统计学意义(P<0.05);5-FU组和NS组比较,差异亦有统计学意义(P<0.05);而不同剂量实验组之间分别进行两两比较,其差异均无统计学意义(P>0.05)。白细胞计数、血小板计数及血红蛋白计数三项指标两两比较中,不同剂量实验组与NS组比较其差异无统计学意义(P>0.05),而与5-FU组比较差异有统计学意义(P<0.05);5-FU组和NS组比较,差异亦有统计学意义(P<0.05);不同剂量实验组之间分别进行两两比较,其差异均无统计学意义(P>0.05);红细胞计数两两比较中,差异均无统计学意义(P>0.05)。结论:中人氟安瘤内植入对人肝癌裸鼠异位移植瘤的抑瘤作用明显,在有效剂量范围内全身毒性反应不明显。

Inhibitory effect of fluorouracil-implant on heterotopic transplanted tumor of human hepatocellular carcinoma in nude mouse

Abstract:Objective:To study the antitumor effect of fluorouracil-implant on heterotopic transplanted tumor of human hepatooellular carcinoma and the relevant mechanism.Methods:50 nude mice bearing heterotopic transplant of human HCC were randomly divided into 5 groups:negative control group(NS group),positive control group 5-FU group,fluorouracil-implant(100mg/kg) group,(200mg/kg) group,and(300mg/kg) group.Drugs were injected into the tumors The effect on tumor growth was evaluated by measuring tumor size prior and post treatment for 15 days and the tumor growth curve was mapped.After 15 days’ treatment,the nude mice were executed and tumors were dissected.Then the tumors were weighed to calculate the inhibition rate.The growth and the change of BW of nude mice before and after the experiment were observed and blood samples were collected for blood routine.Results:All the nude mice in fluorouracil-implant(300mg/kg) group were dead during the first week,so they were not included in the statistical analysis.The average tumor size of NS group,5-FU group,fluorouracil-implant(100mg/kg),and(200mg/kg) group were(842.78±100.15),(408.25±40.68),(313.12±18.53),and(171.74±10.09) mm3 respectively.The tumor inhibition rate of Fluorouracil-Implant(200mg/kg) group was 73.5%,was significantly higher than than of NS group(P<0.01),and 5-FU group(P<0.05).The tumor inhibition rate of Fluorouracil-Implant(100mg/kg) group was 52.4%,with no statistical significance compared with 5-FU group(P<0.05).Comparison between fluorouracil-implant(200mg/kg) and(100mg/kg) group there was statistical significance(P<0.05).Body weight and weight gain of nude mice,had no significant difference between Fluorouracil-Implants groups and NS group(P<0.05);however,there was statistical significance between Fluorouracil-Implant groups and 5-FU group,and between 5-FU group and NS group(P<0.05);but not between the two Fluorouracil-Implant groups(P>0.05).The differences of the count of WBC,PLT and HGB between Fluorouracil-Implants groups and the NS group showed no statistical significance(P>0.05),but was observed between Fluorouracil-Implant groups and 5-FUgroup(P<0.05) and between 5-FUgroup and NS group(P<0.05).Comparisons of the count of RBC among these groaps showed no statistical significance(P>0.05).Conclusion:The injection of Fluorouracil-Implant into tumor can obviously inhibit the growth of heterotopic transplanted tumor of human hepatooellular carcinoma,and the systemic toxicity is not obvious within the effective dose range.
