| 摘 要： | 目的:寻找一种适用于Western boltting实验的提取人心房组织膜蛋白的组织裂解液的配制方法。方法:配制两种不同的组织裂解液A、B。应用组织裂解液A、B及Bio-Rad公司提取膜蛋白的试剂盒,分别对人右心耳组织进行膜蛋白提取,聚丙烯酰胺凝胶电泳(SDS-PAGE)对蛋白进行分离,免疫印迹法检测SK2膜蛋白的表达,比较三种裂解方法的裂解效果。结果:三种不同的裂解方法提取的膜蛋白,其中用组织裂解液A和组织裂解液B提取的膜蛋白能满足Western blotting的实验要求,比用试剂盒提取的方法实验效果好。结论:组织裂解液A和组织裂解液B可作为人心房组织提取SK2膜蛋白的组织裂解液。

| Abstract: | Objective:To search a tissue lysate for the extraction of membrane protein from human atrial tissue.Methods:The membrane protein was extracted from human right atria tissue by three different tissue lysates.The total protein was collected and separated by SDS-PAGE,and then the membrane protein SK2 was detected by Western blotting.Results: Lysis buffer A and B could meet the requirements of Western blotting experiment.The results of lysis buffer were better than that of the kit in extracting membrane protein SK2 from human atrial tissue.Conclusion: The lysis buffer A and B are more appropriate for extracting membrane protein from human atrial tissue. |
