4NQO饮水法诱发小鼠头颈部鳞状细胞癌模型的建立

摘    要:目的快速建立头颈部鳞状细胞癌(简称鳞癌)动物模型。方法Balb/c小鼠65只,随机分为A、B、C、D 4组,分别有20、20、20、5只小鼠,A、B、C 3组为实验组,分别用0.002%、0.005%、0.01%的4-硝基喹啉-1-氧化物(4-nitroquinoline-1-oxide,4NQO)饮水喂养,D组为对照组饮用自来水。肉眼及组织学观察比较4组小鼠癌变过程。结果A、B、C各组分别于第16、11、9周出现舌背黏膜异常增生。到32周A组小鼠舌黏膜中重度异常增生53.8%,原位癌23.1%,浸润癌23.1%;B组小鼠舌黏膜重度异常增生8.3%,原位癌1 6.7%,浸润癌75%;C组小鼠舌黏膜原位癌8.3%,浸润癌91.7%;D组小鼠均无舌黏膜病变。随饮用4NQO剂量增加,小鼠舌背黏膜异常增生程度越重(P〈0.01)。结论4NQO饮水法诱发小鼠头颈部鳞癌,组织病理学特征与人头颈部鳞癌相似,随致癌剂作用时间延长和4NQO浓度的增加,小鼠舌黏膜异常增生越严重。0.01%的4NQO饮用23~32周是建立头颈部浸润鳞癌动物模型的适宜方法。

Abstract:OBJECTIVE To rapidly establish a mouse model with head and neck squamous cell carcinoma.METHODS 65 Balb/c mice were randomized into 4 groups(A,B,C and D).Animals in group A,B and C(each contained 20 mice) were fed with 0.002%,0.005%and 0.01%4NQO solution respectively.Group D was the control group,which had 5 mice and the animals were fed with tap water.
