| 摘 要： | 将生物素—亲和素放大系统(BAS)与McAb ELISA 及FA 技术结合,建立ABC-McAb 和BA-FA 技术,并与组织培养相结合初步应用于眼科临床检测HSV 抗原。实验表明ABC-McAb ELISA 的灵敏度比McAb ELISA 高2.5～4倍,特异性较好,与其它感染眼部的病毒无交叉反应,与病毒分离符合率达96%,特异性95%;BA-FA 技术可直接进行临床标本检测,与病毒分离阳性一致率达91%,该技术与组织培养相结合可明显提高培养24小时阳性检出率。

| Abstract: | The ABC-McAb ELISA and BA-FA techniques were established for rapid detection ofherpes simplex virus(HSV)from the patients presumed HSV keratitis.The detection limitof the ABC-McAb ELISA was 2.5-4 times more than that of McAb ELISA.There wasno cross reaction with the Cytomegalovirus,Adenovirus and Chlamydia Trachomatis 55antigens.We detected 50 clinical eye specimens by ABC-McAb ELISA and BA-FA techni-ques and the results were compared with those of tissue culture.The identity of ABC-McAb ELISA and tissue culture was 96%.The identity of BA-FA and tissue culture was91%.This method provided a more sensitive and specific test for detection of HSV inclinical specimens and could increase the positive ratio of the tissue culture detection ofHSV within 24 hours. |
