RhoC基因高表达对胃癌细胞血管内皮细胞生长因子、碱性成纤维细胞生长因子表达的影响

摘    要:目的 探讨RhoC基因对胃癌细胞血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)表达的影响及其意义。方法 常规脂质体转染法将RhoC基因重组质粒转染人胃癌细胞系SGC7901;应用免疫细胞化学链霉菌抗生物素蛋白-过氧化酶(SP)法检测胃癌细胞转染RhoC基因重组质粒前后VEGF、bFGF蛋白表达的变化。结果 转染RhoC基因重组质粒在SGC7901细胞中有稳定表达。转染RhoC基因重组质粒SGC7901表达VEGF、bFGF明显强于对照组。结论 体外条件下RhoC基因可促进胃癌细胞VEGF、bFGF的表达。这可能是RhoC基因促进胃癌细胞侵袭、转移的分子基础。RhoC基因的表达可望作为估计胃癌转移的参考指标。

Abstract:Objective To explore the influence of RhoC gene on the expression of vascular en- dothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in human gastric cancer cell line. Methods The recombinant vector (pcDNA 3.1-RhoC) was transfected into SGC7901 cells with LIPFECTAMINE~(TM)Reagent. The expression of VEGF and bFGF was detected by the immunohistoche- mical stain in SGC7901 cells. Results The recmbinant vector (pcDNA 3.1-RhoC) was expressed steadi- ly in SGC7901 cells. The expression of VEGF and bFGF in the pcDNA 3.1-RhoC-transfected SGC7901 cells was significantly stronger than that of the pcDNA 3.1-transfected SGC7901 cells and untransfected SGC7901 cells. Conclusion RhoC gene could increase the expression of VEGF and bFGF in human gas- tric cancer cell line in vitro, which might be contributed to the molecular foundation of RhoC gene en- hancing invasiveness and metastasis of human gastric cancer cell line. The expression of RhoC gene may act as a good marker for the metastasis of gastric cancer.
