催乳素保护人B淋巴细胞免其凋亡的实验研究

目的:研究人催乳素(PRL)在B淋巴母细胞系IM-9细胞凋亡中的保护作用。方法:在地塞米松诱导IM-9细胞凋亡的实验体系中加入PRL(10ng/ml),DNA琼脂糖凝胶电泳观察PRL对地塞米松诱导IM-9细胞凋亡的保护。结果:地塞米松组诱导的IM-9细胞在48h后出现细胞凋亡,DNA琼脂糖凝胶电泳显示有大量小分子DNA片段形成,呈典型的细胞凋亡DNA梯形带,地塞米松-PRL组的IM-9细胞的DNA经电泳后未出现凋亡的DNA条带。结论:地塞米松能诱导IM-9细胞的凋亡,PRL能保护IM-9细胞,免其凋亡。

The experimental study on the protective effect of PRL on human B lymphocytes against apoptosis

Objective: To study the protective effect of PRL on human B lymphocytes against the apoptosis. Methods: PRL (10ng/ml) was added to the experiment of cell apoptosis induced by Dexamethasone. Then by DNA Agarose gel electrophoresis, the protective effect of PRL on IM-9 cells against apoptosis was observed. Results: IM-9 cells induced by Dexamethasone showed apoptosis after forty-eight hours,whose DNA showed a number of DNA fragments in the Agarose gel electrophoresis containing EB, which looked like the typical DNA ladder echelon. The group given Dexamethasone and PRL didn’t show DNA echelon of apoptosis. Conclusion: Dexamethasone could induce apoptosis of IM-9 cells, but the addition of the PRL could protect IM-9 cells against apoptosis.
