人肝癌组织特异性γ-谷氨酰转移酶分离纯化

摘    要:目的:分离纯化人肝癌组织中特异性γ-谷氨酰转移酶(GGT),为进一步制备其抗血清提供抗原材料,以便对肝癌早期诊断进行研究。方法:人肝癌组织经匀浆、离心、溶解、透析、凝胶层析及凝集素亲和层析等步骤进行分离纯化;纯化过程用IFCC推荐方法监测GGT催化活性,紫外法监测蛋白质出峰图谱,Lowry法测定蛋白含量。结果:经过粗提、DEAE阴离子交换层析、凝胶过滤层析及凝集素亲和层析等纯化步骤得到电泳纯的GGT,其比活为10.58U/mg蛋白,纯化倍数为84.9,得率为1.60%。结论:该方法能够较好地分离纯化人肝癌组织中特异性GGT。

Abstract:Objective:To Purify special γ-glutamyltraspeptidese from human liver cancer tissue, which is then used to produce antibodies targeted for early diagnosis of liver cancer. Methods:Human liver cancer tissue is purified by rough extraction, dialyzation, ion-exchange chromatography and lectin affinity chromatography etc. Results:The special GGT in human liver cancer tissue was purified. The catalysis units per milligram protein of GGT have been increased about 84.9 times. And the yield of the GGT was 1.60%. Conclusion:This protocol is efficient to purity GGT from human liver cancer tissue.
