奥沙利铂、氟脲嘧啶及甲酰四氢叶酸治疗晚期大肠癌

目的:观察奥沙利铂(oxaliplatin,L-OHP)联合氟脲嘧啶(5-Fu)及甲酰四氢叶酸(CF)治疗大肠癌的疗效和毒副反应。方法:L-OHP120mg/m2持续2h静脉滴入,d1;CF100mg/m2静脉滴入d1~d5,5-Fu375mg/m2静脉滴入,d1~d5.21d重复。结果:52例患者中,完全缓解(CR)2例,部分缓解(PR)22例,稳定(NC)18例,进展(PD)10例,有效率(CR PR)54%。毒副反应可耐受,主要表现外周神感觉异常,症状可逆。结论:奥沙利铂联合氟脲嘧啶及甲酰四氢叶酸治疗大肠癌可以获得比较高的疗效,显著提高患者生活质量。

Clinical study of oxaliplatin in treatment of advanced colorectal carcinoma

Objective:To evaluate the efficacy and adverse of oxalipatin combination with fluorouracil and leucovorin in the treatment of advanced colorectal carcinoma.Methods:Oxalilatin was administered as a 2-hour infusion at a dose of 120mg/m2 on 1 day;leucovor 100mg/m2 on day 1 to 5;fluorouracil 375mg/m2 n day 1 to 5.The schedule was repeated every 21 days,with at least two cycles.Results:fifty-two patients were evaluable for treatment outcome.two obtobtained CR,22 PR,18 NC,10 PD,and the overall response rate was 54%.A dose-related and reversible peripheral sensory neuropathy was the dose-limiting toxicity.The adverse reaction was tolerable.Conclusions:The oxaliplatin combination with fluorouracil and leucovorin is a highly eeffective regimen for patients with advanced gastric carcinoma,with a manageable toxicity.
