右归饮对老年肾虚证小鼠影响的蛋白质组学研究

摘    要:目的:从蛋白质表达水平观察中药方剂右归饮对老年小鼠生理性肾虚证动物模型的影响。方法:将昆明系雄性小鼠随机分为老年肾虚证模型组、右归饮(5.6 g.kg-1,1次/d连续ig 2个月)组与青年正常对照组3组,对其肝、肾、睾丸组织蛋白质提取液及血清分别进行蛋白质双向电泳并对电泳图谱进行分析,找出右归饮调节纠正的蛋白质,进行质谱分析鉴定和Western-blot验证。结果:找到右归饮调节纠正的蛋白质点22个,质谱鉴定出17种蛋白质。结论:找到并鉴定了右归饮对老年小鼠生理性肾虚证动物模型调节纠正的17种蛋白质。

Abstract:Objective : To observe the effects of Youguiyin decoction on the mice with Kidney-Deficiency Syndrome in protein expression level. Method : Kunming male mice were randomly divided into three groups as follows: the kidney-deficiency model group, the Youguiyin group (5.6 g·kg-1 daily for 2 months)and the youth normal group. The proteins in liver,kidney,testis tissue and serum of the mice were separated and analysed by two-dimensional gel (2-DE) to find the proteins regulated and corrected by Youguiyin. These proteins were identified by mass spectrograph. Result : Tenty two regulated and corrected protein spots were detected, and 17 proteins spots were identified. Conclusion : This paper screens and identifies 17 proteins regulated and corrected by Youguiyin decoction.
