| 摘 要： | 目的研究各种因素对神经生长因子(NGF)生物活性及免疫原性的影响。方法把天然NGF在不同浓度的变性剂、去污剂、氧化还原剂及其配伍中或在不同pH值、不同温度下进行孵育处理,测定其生物活性和免疫原性的变化。结果NGF在8mol/L尿素中孵育过夜后生物活性下降10倍,在浓度低于5mol/L的尿素中孵育过夜后生物活性基本不变;十二烷基硫酸钠和巯基乙醇对NGF生物活性破坏较大,且尿素可增加巯基乙醇的破坏作用;还原型谷胱甘肽和氧化型谷胱甘肽配合使用对NGF生物活性的影响较小;NGF对温度敏感,在60℃孵育1h后活性降低约30倍,在37℃孵育48h生物活性开始下降;NGF在酸、碱中稳定。生物活性降低的NGF免疫原性未见明显改变。结论NGF的生物活性易受多种因素影响,但其免疫原性较稳定。
