育泡饮对大鼠卵巢颗粒细胞间隙连接蛋白43表达及分泌功能的影响

摘    要:[目的]观察育泡饮对体外培养大鼠卵巢颗粒细胞(GC)间隙连接蛋白43(CX43)表达及雌二醇(E2)分泌的影响,探索中药促进卵泡发育的可能作用机制。[方法]制备大鼠卵巢颗粒细胞进行原代培养,细胞培养48 h后,加入促卵泡激素(FSH)50 ng/mL和不同剂量的育泡饮,继续培养48 h,放免法测定颗粒细胞培养液中E2的含量,Westernblot法检测CX43蛋白的表达。[结果]1)育泡饮能够促进卵巢颗粒细胞分泌E2,随着药物浓度的增大,E2分泌逐渐增加。2)育泡饮可明显促进颗粒细胞CX43蛋白的表达,其中高剂量组的促进作用与FSH组作用相似。[结论]育泡饮能明显促进卵巢颗粒细胞CX43蛋白的表达及E2分泌,说明育泡饮可通过上调CX43蛋白的表达促进颗粒细胞的增殖,增强颗粒细胞的抗凋亡能力,进而调节卵泡的发育。

Abstract:[Objective] To assess the effects of Yupao Yin on expression of CX43 in ovarian granulosa cells(GC) of rats and secretion quantity of E2. [Methods] Granulosa cells are isolated from rats ovaries and cultured for 48 hours, followed by culture in medium system containing FSH (50 ng/mL) and different dosage of Yupao Yin for 48 hours. The expression of CX43 is measured by western blot and the secretion quantity of E2 is performed by radioimmunoassay. [Result] Yupao Yin can promote secretion of E2 and the secretion quantity of E2 gradually increases with the increase of the dosage. Yu pao Yin can significantly stimulate the expression of CX43. Amomg them the effect of high dosage group is similar with FSH group. [Conclusion] Yupao Yin obviously promote the expression of CX43 and secretion of E2 of ovarian GC of rats. It is showed that Yupao Yin can promote proliferation of GC and enhance the ability of GC anti-apoptosis through up-regulation the expression CX43. And then to promote the development of follicular.
