摘    要:RNA干扰作用(RNA i)是通过双链RNA(dsRNA)引发的转录后基因沉默机制。RNA i主要是指dsRNA分子进入细胞内,特异性降解与之同源的mRNA,从而特异高效地抑制相应基因表达活性的现象。近年来,RNA i以其高特异性、高效性等显著优势已成为研究基因功能的新手段,在功能基因组学、基因表达调控机制研究等领域得到了广泛的应用。在此对RNA干扰技术的发展历程、作用机制、作用特点及应用前景予以综述。

Abstract:RNA interference(RNAi)is a post-transcriptional gene silencing induced by the introduction of double-stranded RNA(dsRNA)which triggers the degradation of a homologous mRNA.RNAi is a newly developed simple gene-silence tool which is of high specificity and high efficiency,which has been widely used in functional genome and regulatory mechanism of gene expression.This article reviews the development,mechanism of action,characteristics,and application prospect of RNAi.
