摘    要:目的 检测不同牙周状态下牙龈卟啉单胞菌(P.gingivalis)唾液酸酶基因表达的差异,探讨唾液酸酶在慢性牙周炎发生发展中的作用.方法 通过Clustal W将PG0352编码的氨基酸与已知功能的氨基酸做同源比对,明确编码唾液酸酶的基因,选择不同牙周状态下的龈下菌斑样本104例,PCR检测唾液酸酶基因,RT-PCR法比较不同条件下唾液酸酶基因表达的差异.结果 同源比对结果显示PG0352含有3个唾液酸酶基因特有的功能基序.病变重部位、病变轻部位及健康部位P.gingivalis的检出率分别为85.70%、76.19%和40.00%,唾液酸酶基因的检出率分别为91.67%、87.5%和87.5%.慢性牙周炎患者病变重部位唾液酸酶表达水平高于病变轻部位和健康部位(P<0.05).结论 PG0352编码唾液酸酶,P.gingivalis唾液酸酶的表达水平与牙周状态有关,推测P.gingiwlis唾液酸酶可能与慢性牙周炎的发生发展关系密切.

Abstract:Objective To analyze the expression level of sialidase in P.gingivalis in different situations and discuss the role of sialidase in the genesis and progress of chronic periodontitis.Methods Clustal W was used to conduct homology analysis of PG0352 and sialidase gene.A total of 104 subgingival plaque samples were collected in this study.PCR was performed to evaluate the detection rate of sialidase gene,RT-PCR was used to determine the expression level of sialidase gene in P.gingivalis.Results The detection rates of P.gingivalis in lesion site,non-lesion site and healthy people were 85.70%,76.19% and 40.00%,respectively.The detection rates of sialidase gene in lesion site,non-lesion site and healthy people were 91.67%,87.5% and 87.5%,respectively.The expression level of sialidase was higher in lesion site than those in non-lesion site and healthy people.Conclusion PG0352 encodes sialidase.The expression level of sialidase is association with the periodontal situation,suggesting that sialidase is involved in the genesis and progress of chronic periodontitis.
