摘    要:目的 研究胰岛素受体底物1(IRS-1)在小鼠前成骨样MC3T3-E1细胞分化过程中的表达.方法 在小鼠前成骨样MC3T3-E1细胞分化成熟过程中,分别在不同时点收集细胞及上清液.骨钙素含量用放射免疫法测定;碱性磷酸酶活性用α-磷酸奈酚法测定;Ⅰ型胶原、骨涎蛋白、骨桥素和IRS-1的mRNA表达应用半定量RT-PCR检测;免疫印迹法测定IRS-1蛋白水平.结果 在小鼠前成骨样MC3T3-E1细胞分化过程中,IRS-1的表达量逐渐增加,而且与Ⅰ型胶原、骨钙素、骨涎素和骨桥素呈正相关.结论 IRS-1参与MC3T3-E1细胞的增殖、成熟与矿化的全过程.

Abstract:Objective To investigate the expression profile of insulin receptor substrate 1(IRS-1) during the differentiation of preosteoblastic cells.Methods Pnitrophenyl phosphate assay was used to evaluate alkaline phosphatase activity,RIA was used to evaluate osteocalcin.Semi-quantitive RT-PCR was used to examine the mRNA levels of bone sialoprotein(BSP),osteopontin(OPN),collagen type Ⅰ(Col Ⅰ)and IRS-1 in MC3T3-E1 cells.Western blot was adopted to evaluate the level of IRS-1 protein.Results During differentiation of MC3T3-E1 cells,IRS-1 mRNA and protein levels gradually increased.Osteocalcin,Col Ⅰ,OPN and BSP were positively related to IRS-1.Conclusion IRS-1 may be involved in the proliferation,differentiation and mineralization of murine preosteoblastic MC3T3-E1 cells.
