皮质发育障碍伴癫痫发作动物模型的制作

摘    要: 目的建立X线—匹罗卡品双重干预的皮质发育障碍(DCDs)鼠伴癫痫发作模型。方法将6只SD孕鼠随机分为实验组和正常对照组,实验组孕鼠给予X线照射,正常对照组不做任何处理。待仔鼠出生后60 d观察雄性仔鼠行为、脑电图、大脑形态及皮质和海马的病理改变。60 d的雄性仔鼠腹腔注射氯化锂—匹罗卡品诱发癫痫,比较潜伏期、注射药物总剂量、Ⅴ级癫痫发作率以及行为学和自发性癫痫发作情况。结果与正常对照组雄性仔鼠比较,实验组DCDs雄性仔鼠大脑体积变小,可见明显畸形;HE染色显示皮层变薄,神经元排列紊乱,海马CA1、CA2中断,可见团块状结构;未发现自发性癫痫发作,脑电图无异常放电,有活动增多等表现。注射氯化锂—匹罗卡品后实验组DCDs雄性仔鼠癫痫发作潜伏期缩短(P<0.05),用药剂量减少(P<0.05),Ⅴ级发作率增高(P<0.05);癫痫持续发作后易激惹,具有攻击行为,并在静止期及慢性期出现自发性癫痫发作;HE显示皮层厚度减小。结论X线—匹罗卡品双重干预能成功建立弥漫性DCDs模型,并产生自发性癫痫发作。

Establishment of Rat Model with Disorders of Cortical Development and Epilepsy

Abstract:Objective To establish the rat model with disorders of cortical developments(DCDs) and epilepsy induced by lithium chloride and pilocarpine following prenatal X-ray radiation.Methods Six pregnant SD rats were randomly divided into experimental group and control group.The experimental group were given X-ray radiation while the control group not.Sixty days after being born,the male newborn rats were observed in behavior,EEG,brain shape and the pathological changes of cortex and hippocampus.The male newborn rats of the experimental group were injected with lithium chloride and pilocarpine to induce epilepsy.The two groups were compared in terms of incubation period,the total doses of injected drug,Grade V attack rate,behavior and spontaneous seizures.Results Compared with the newborn rats of the control group,those of the experimental group presented with smaller brain and obvious deformity;disordered arrangement of cortical neurons and interrupted CA1 and CA2 in hippocampus with abnormal neuron aggregation,shown by HE;increased activity without spontaneous epilepsy and abnormal discharge in EEG.After the injection of lithium chloride and pilocarpine,the newborn rats of the experimental group had shorter incubation periods(P < 0.05),less doses(P < 0.05) and higher Grade V attack rate(P < 0.05),compared with those of the control group.After recurrent epilepsy,the rats became more irritable and aggressive,and presented with spontaneous seizures during the latent and chronic period and thinner cortex revealed by HE.Conclusion Double intervention by X-ray radiation,lithium chloride and pilocarpine can successfully establish the rat model with diffuse DCDs and result in spontaneous seizures.
