阿魏酸川芎嗪抗血栓形成作用及其对血小板中性粒细胞黏附的影响

摘    要:
目的 观察阿魏酸川芎嗪(LF)的抗血栓作用及其对血小板中性粒细胞黏附的影响,并探讨其可能机制.方法 采用电刺激大鼠颈动脉血栓形成法和动静脉旁路法血栓形成法评价LF对血栓形成的对抗作用;运用玫瑰花结黏附试验观察LF对血小板中性粒细胞黏附的影响;采用流式细胞术法检测LF对血小板P-选择素(CD62p)表达的影响.结果 LF能明显抑制大鼠电刺激颈动脉法和动静脉旁路法引起的血栓形成;减少CD62p的表达;抑制血小板与中性粒细胞的黏附,其半数抑制浓度(IC50)为64.3μmol· L-1.结论 LF具有明显的抗血栓形成作用,减少CD62p的表达,从而阻抑血小板中性粒细胞黏附有关.

Abstract:
Objective To investigate the antithrombotic effects of ligustrazine ferulate and its possible mechanisms.Methods The method of electrically stimulated carotid thrombosis and the method of bypass thrombosis between carotid and jugular vein in rats were used to evaluate the antithrombotic effects of ligustrazine ferulate;rosette assay method was performed to observe platelet-neutrophi1 interactions;platelet P-selectin(CD62p)was detected using flow cytometry.Results Ligustrazine ferulate not only protected against thrombosis caused by electrical stimulation and carotid and jugular bypass,but also reduced the expression of platelet P-selectin,and decreased the binding of thrombin stimulated platelets to neutrophils.The half inhibitory concentration(IC50) was 64.3 μmol·L-1.Conclusion Our findings showed that ligustrazine ferulate had evident antithrombotic effects,and the mechanisms could be intimately related to reduced expression of platelet P-selectin and suppression of platelet neutrophil interactions.
