RAGE基因型在2型糖尿病伴慢性牙周炎患者颊黏膜中的检测

摘    要:目的 对2型糖尿病伴慢性牙周炎患者颊黏膜中糖基化终产物受体(RAGE)进行检测,探讨其可能致病位点基因型.方法 分别选取2型糖尿病伴慢性牙周炎患者(DMCP组)、单纯慢性牙周炎患者(CP组)和健康对照组(H组)各50例.采集颊拭子,进行牙周检查,对比牙周状况.从样本中提取DNA,RAGE PCR产物检测,PCR质量控制,限制性内切酶酶切产物,确定可能的基因型.结果 RAGE基因中-429T/C位点和2184A/G位点在DMCP组、CP组和H组中无统计学差异(P>0.05),而1704G/T位点GG基因型在DMCP组和CP组的表达要高于H组,差异有统计学意义(P<0.05).结论 RAGE基因中内含子区域1704G/T位点GG基因型在DMCP组和CP组中出现频率高.2型糖尿病伴慢性牙周炎和单纯慢性牙周炎可能与1704G/T位点GG基因型表达有关.

Abstract:Objective To detect the receptor for advanced glycation end products(RAGE)in buccal mucosas of type 2 diabetic patients with periodontitis,and investigate the pathogenic site.Methods Totally 50 type 2 diabetic patients with periodontitis(DMCP),50 chronic periodontitis patient(s CP)and 50 healthy(H)were selected for the study.Bucca swabs from every subject were collected,and their periodontal situations were examined.DNA were extracted from the collected samples,and the PCR products of RAGE were detected by electrophoresis to control the products quality.Results There was no statistic difference among the groups-429T/C site and 2184A/G site(P > 0.05).1704G/T site GG genotype showed a higher frequency in group DMCP and group CP than in group H(P < 0.05).Conclusion In the receptor for advanced glycation end products gene,we detected 1704G/T site GG genotype in intron region.This site showed a high frequency in group DMCP,while there was no statistic difference between group CP and group H.The pathogenic site of DMCP and CP might be connected with 1704G/T GG genotype.
