摘    要:  目的 建立一种稳定的大鼠尾核神经元的分离和体外培养方法,用于中枢神经元离子通道特性的研究。方法 自新生24 h内的Wistar乳鼠大脑中分离出尾核,结合酶消化及机械吹打分离尾核神经元,进行体外原代神经元培养。采用全细胞膜片钳技术记录电压门控钠通道电流和电压门控钾通道(Kv)电流。结果 分离出的神经元形态正常,细胞膜光滑有弹性,保存了主要离子通道的活性。结论 本方法适合用于中枢神经系统神经元离子通道的研究。

Abstract:Objective To establish a reliable method for primary culture of neurons from caudate nucleus of rodent,and study the characteristics of ion channels by using whole cell patch-clamp technique.Methods Primary neurons from caudate nucleus of newborn Wistar rats(within 24 h after birth) were isolated by using enzyme digestion and mechanical trituration.The neurons were then used in the analysis of both voltage-gated sodium currents and voltage-gated potassium currents using the whole cell patch-clamp technique.Results Primary neurons with high quality could be obtained from caudate nucleus,as indicated by the smooth membrane,elasticity and the activities of primary iron channels.Conclusion This method is suitable for the study of the characteristics of ion channels.
