血红素氧合酶-1对大鼠主动脉平滑肌细胞增殖的影响

摘    要:目的探讨血红素氧合酶-1(HO-1)蛋白表达对大鼠主动脉平滑肌细胞(RASMC)增殖的影响。方法大鼠主动脉平滑肌细胞株经复苏、传代培养后用于实验,设置空白对照组、HO-1诱导组、HO-1抑制组,后两组分别加用血晶素、锌原卟啉Ⅸ与细胞共同孵育,用Western blot法检测HO-1表达量,并检测HO-1活力;用MTT法检测RASMC增殖能力。同时,还观察了血晶素、锌原卟啉Ⅸ的细胞毒性作用。结果上调HO-1蛋白表达及升高HO-1活力能显著抑制血清刺激的RASMC增殖,反之,抑制HO-1蛋白表达及其活力则增强血清刺激的RASMC增殖。实验浓度的血晶素、锌原卟啉Ⅸ无明显细胞毒性作用。结论 HO-1蛋白高表达能有效抑制RASMC增殖。

The Antiproliferation Effects of Heme Oxygenase-1 on Rat Aortic Smooth Muscle Cells

Abstract:Objective To explore the antiproliferation effects of heme oxygenase-1(HO-1) protein expression on rat aortic smooth muscle cells(RASMC).Methods RASMC were revived and cultured and divided into control groups,HO-1 induced group and HO-1 depressed group.cells in HO-1 induced group and HO-1 depressed group were added respectively with hemin and zinc protoporphyrin Ⅸ(ZnPPⅨ) to an end-point concentration of 10 μmol/mL.The HO-1 protein expression was detected by western blot analysis and the HO-1 protein activity was also measured.The cell proliferation activity was evaluated by MTT assay.The cytotoxicity of hemin and ZnPPⅨ was measured by Lactate Dehydrgenase colorimetry and Trypan blue exclusion tests.Results Hemin could up-regulate the HO-1 protein expression and its activity,significantly inhibit the serum-stimulated proliferation in RASMC.On the contrary,ZnPPⅨ decressed the HO-1 protein expression and its activity,significantly promoted the serum-stimulated proliferation in RASMC.Both hemin and ZnPPⅨ in the experimental concentration exhibited no apparent cytotoxocity.Conclusion HO-1 protein overexpression could surppress the proliferation of RASMC effectively.
