美金刚对急性氧乐果中毒的实验治疗研究

摘    要:目的研究美金刚(MEM)对急性氧乐果中毒的疗效。方法(1)小鼠用氧乐果(20 mg/kg)灌胃染毒后,经腹腔注射生理盐水(未治疗组)、10 mg/kg MEM(MEM治疗组)或15 mg/kg阿托品(At治疗组),观察3组动物中毒表现和死亡结局。(2)大鼠用氧乐果(40 mg/kg)灌胃染毒后,经腹腔注射生理盐水(未治疗组)或18 mg/kgMEM(MEM治疗组),观察两组动物的中毒表现和死亡结局。结果(1)小鼠MEM治疗组惊厥和呼吸困难发生率均较未治疗组减少,肌颤出现的时间延后,程度减轻,存活率(70%)明显高于未治疗组(10%),差异有统计学意义(P<0.05);(2)大鼠MEM治疗组惊厥、呼吸困难和肌颤出现时间延迟,肌颤程度减轻,存活率(70%)明显高于未治疗组(20%),差异有统计学意义(P<0.05)。结论MEM对急性氧乐果中毒有一定的治疗作用。

Abstract:Objective To study the therapeutic efficacy of memantine(MEM)against acute toxicity of omethoate.Method(1)Soon after omethoate(20 mg/kg)were given to mice by gastrogavage,physiological saline,MEM(10 mg/kg)or atropine(15 mg/kg)were administered to three groups by intraperitoneal injection.Toxic signs and survival rate were recorded.(2)Soon after omethoate(40 mg/kg)were given to rats by gastrogavage,physiological saline or MEM(18 mg/kg)were intraperitoneally injected to two groups.Toxic signs and survival rate were recorded.Result(1)The decreased incidence rate of convulsion and dyspnea,the alleviated intensity and delayed onset of muscular fasciculation were seen in omethoate-exposed mice after MEM treatment as compared with non-treatment group.Survival rate was significantly higher in MEM treatment group than in non-treatment group(P<0.05).(2)The onset of convulsion,dyspnea and muscular fasciculation were delayed and the survival rate was significantly increased in rats of MEM treatment group compared with non-treatment group.Conclusion MEM has therapeutic efficacy against acute toxicity of omethoate.
