蟾蜍灵诱导人急性淋巴白血病细胞凋亡的实验研究

摘    要:目的研究蟾蜍灵对人急性淋巴白血病(CEM)细胞的作用。方法台盼蓝拒染法观察蟾蜍灵对细胞增殖的抑制作用;流式细胞术检测蟾蜍灵作用后细胞凋亡情况。结果10~80nmol/L蟾蜍灵于24h开始明显抑制CEM细胞生长(P<0.01)。24,48和72h抑制细胞增殖50%的药物浓度分别为15.13,9.59和6.57nmol/L;20nmol/L,40nmol/L蟾蜍灵作用24h后出现典型的细胞凋亡形态学改变;20nmol/L蟾蜍灵处理12,24和48h凋亡细胞百分率分别为2.46%、17.92%和66.26%;40nmol/L蟾蜍灵处理12,24和48h凋亡细胞百分率分别为5.19%、32.71%和80%以上;凋亡细胞百分率呈剂量和时间依赖性。结论蟾蜍灵以时间、剂量依赖方式抑制CEM细胞生长、诱导凋亡。

Abstract:Objective To investigate the effect of bufalin on inducing cell apoptosis in human acute lymphoblastic cell line CEM. Methods Cell viability was determined by trypan blue dye exclusion. Apoptosis was evaluated by morphology and flow cytometry. Results Cell viability was decreased significantly after treatment with 10nmol/L bufalin for 24 h (P < 0.01). Proliferation was inhibited in a concentration- and time-dependent manner. The 50% inhibiting concentration (IC50) at 24 h,48 h and 72 h were about 15.13 nmol/L,9.59 nmol/L,and 6.57 nmol/L,respectively. Apoptotic changes of morphology was observed 24 hours or more after treated by 20 nmol/L of bufalin or highera,including cell shrinkage,chromatin agglutination and formation of apoptotic corpuscular.The apoptotic percentage was 2.46%,17.92% and 66.26%,respectively after treatment with 20 nmol/L bufalin for 12 h,24 h and 48 h. The apoptotic percentage was 5.19%,32.71% and more than 80%,respectively after treatment with 40 nmol/Lbufalin for 12 h,24 h and 48 h. The apoptotic percentage was significantly increased in time- and dose-dependent manner. Conclusion Bufalin could effectively inhibit the growth and induce the apoptosis of CEM cells in a concentration- and time- dependent manner.
