三七总皂甙对大鼠肝脏短期缺血再灌注损伤早期钙离子含量变化的影响

摘    要:目的 ]观察三七总皂甙对大鼠肝脏短期缺血再灌注损伤早期线粒体钙离子含量的影响 .方法 ]通过直接注入并保留药物的方法 ,用原子吸收分光光度法测定大鼠肝脏局部缺血再灌注损伤模型肝细胞线粒体与胞浆中的钙离子含量 ,同步观察电子显微镜下组织学改变 .结果 ]用三七总皂甙处理的大鼠肝细胞线粒体钙离子含量在再灌注早期虽有波动 ,但无显著性意义 ;再灌注 90min ,三七总皂甙处理组的大鼠肝细胞线粒体钙离子含量 (6 30± 0 71μg/g)明显低于对照组 (8 73± 2 0 3μg/g) ,缺血再灌注大鼠肝细胞胞浆中钙离子含量在三七总皂甙组与对照组均随着再灌注时间的延长逐渐增加 ;对照组到再灌注 90min时钙离子含量 (14 2 6± 2 6 9μg/g)显著升高 ,而相比之下三七总皂甙组钙离子含量的增加相对比较平稳 ,且各时间组均略低于对照组 ,但未见显著性差异 .电子显微镜下观察到三七总皂甙组的线粒体等超微结构的改变较对照组轻 .结论 ]大鼠肝脏短期缺血再灌注损伤早期线粒体与胞浆中的钙离子含量随着再灌注时间的延长而逐渐增高 ,以胞浆中的变化为甚 ,而超微结构的损伤则以线粒体为主 ;三七总皂甙对大鼠肝脏短期缺血再灌注损伤早期线粒体钙离子含量的影响较对胞浆明显 ;三七总皂甙对大鼠肝脏短期缺血再灌注损伤具有

Abstract:OBJECTIVE To study the effects of the panax notoginseng saponins for calcium content of mitochondria in early stage of short-term ischemia-reperfusion injury in rats. METHODS The calcium content of hepatocyte mitochondria in the model of early stage of short-term ischemia-reperfusion injury in local liver of rats was detected by the method of directly infusing and preserving medicine and atomic absorption spectrophotometer, and the histological changes were synchronously observed by electronic microscope. RESULTS The calcium content of mitochondria in the early stage of short-term ischemia-reperfusion injury in the liver of rats had a little fluctuation, but there was no significant between panax notoginseng saponins group and control group. After reperfusion for 90 mins, the calcium content(6 30 μg/g±0 71 μg/g) in panax notoginseng saponins was much less than that(8 73 μg/g±2 03 μg/g) of control group, one of hepatocyte cytoplasm in both groups increased with prolongation of reperfusional time and one(14 26 μg/g±2 69 μg/g) in control group increased significantly, however panax notoginseng saponins group became stable and each group are slightly lower than control group correspondingly. It was observed under electronic microscope that the lesions of mitochondria in panax notoginseng saponins group were slighter than that in control group. CONCLUSION Calcium of hepatocyte mitochondrion and cytoplasm in early stage of ischemia-reperfusion injury in rats increases with prolongation of reperfusion time, especially in cytoplasm, but the ultrastructive lesion is mainly on mitochondrion. Effects of panax notoginseng saponins for the mitochondrion calcium in early stage of ischemia-reperfusion injury in rat liver are much more obvious. The panax notoginseng saponins has protective function for ischemia-reperfusion injury in rat liver by blocking the inflow of cytoplasm free calcium to mitchondrion.
