摘    要:目的 观察不同发育阶段黑胸大蠊水提取物对离体淋巴细胞增殖的影响. 方法 提取小鼠脾细胞,制成一定浓度细胞悬液.实验分3组:对照组、伴刀豆球蛋白(Con A)阳性对照组和实验组.实验组设卵鞘、若虫、雌虫和雄虫等4组水提取物.每组设低、中、高剂量组,浓度依次为2.5 μg/ml、5.0μg/ml和10.0μg/ml,对照组加相同浓度的完全培养基.在一定条件下进行培养,再加入四甲基偶氮唑盐(methyl thiazolyl tetrazolium,MTT),测出各实验组570 nm波长的吸光度A值,运用方差分析进行统计学处理,以此判断淋巴细胞增殖情况;用吉氏染色法对各组淋巴细胞形态进行观察,通过其细胞体积、胞核及胞浆等的变化判断淋巴细胞增殖情况. 结果卵鞘、若虫、雌虫和雄虫的水提取物中高剂量组的,A值均高于对照组,若虫中剂量组和雌虫中、高剂量组A值依次为0.362±0.033、0.333±0.088、0.354±0.039,与对照组(0.231±0.020)比较,差异有统计学意义(P<0.05),低剂量组内各实验组与对照组之间差异无统计学意义(P>0.05);细胞形态学改变与A值变化一致. 结论黑胸大蠊若虫和雌虫水提取物在一定浓度时有促进离体淋巴细胞增殖的作用.

Abstract:Objective To observe the effect of aqueous extracts (E-Pf) from Periplaneta fuliginosa at different developmental stage on the lymphocyte proliferation in vitro. Methods The suspensions of mouse splenocyte were prepared and distributed into 3 groups: the control, the Con A treated positive control and the experiment (i.e. the E-Pf-treated ) groups, in which the experiment group was further divided into 4 sub-groups : the egg, the nymph, the male adult and the female adult E-Pf. The splenocyte suspension in each sub-group was cultured with high ( 10.0 μg/ml) , medium (5.0 μg/ml) and low ( 2.5 μg/ml) doses of its corre-sponding aqueous extract, respectively. The splenocyte suspension in the control group, however, was cultured merely with complete medium. Methyl thiazolyl tetrazolium (MTT) was then added into all groups, followed by the detection of optical density (A) values of MTT in each group. The proliferations of lymphocytes were judged by the statistical study of variance analysis. Morphological observations on the changes of cell volume, nucleus and cytoplasma were also done after Giemsa staining. Results The A values of all the middle- and high-dose E-Pf-treated subgroups were higher than that of the control group. The A values of the medium-dose nymph E-Pf ( 0.362 ± 0.033 ), the medium-dose (0.333 ± 0.088 ) and high-dose (0.354 ± 0.039 ) female adult E-Pf sub-groups were significantly higher than that of the control group (0.231 ± 0.020) ( P < 0.05 ). No significant differences in A values were found between each of the low-dose E-Pf-treated subgroups and the control group ( P > 0.05 ). The morphological changes of lymphocytes were consistent with the changes of A values. Con-clusion The Periplaneta fuliginosa aqueous extracts of nymph and adult females promote the lymphocyte prolif-eration in vitro.
