IGFBP-1,IGFBP-2在围着床期大鼠子宫内膜中的表达及其与孕激素的关系

摘    要:目的 探讨 IGFBP- 1和 IGFBP- 2在围着床期大鼠子宫内膜中的表达及其与孕激素 (P)的关系。方法 采用放射免疫法测定孕 3~ 10天大鼠血清 P水平 ;用免疫组化 L s AB法及计算机图像分析法检测 IGFBP-1、IGFBP- 2及孕激素受体 (PR)在孕早期大鼠子宫内膜及胚胎滋养层细胞中的表达。结果 孕鼠血清 P呈峰值变化 ,最大值出现在孕 5~ 7天。 PR在大鼠内膜腺、腔上皮 ,基质细胞 ,蜕膜细胞及滋养层细胞中均有表达 ,其在子宫内膜的表达强度也呈峰值变化 ,最大值出现在孕 5~ 7天 ,并与血清 P水平呈正相关 ;而 PR在大鼠滋养层细胞中的表达随孕天升高 ,且与血清 P无相关。 IGFBP- 1和 IGFBP- 2在孕鼠子宫内膜的表达与 PR具有相同细胞定位 ,但在滋养层细胞中没有表达。 IGFBP- 1表达的变化趋势与 PR类似 ,而 IGFBP- 2的表达则是在妊娠后升高并维持相对恒定 ,孕 8~ 9天降到动情期水平 ,孕 10天又迅速升高。二者在围着床期大鼠子宫内膜中的表达与血清 P都呈正相关 ,(r分别为 0 .375和 0 .345 ,P均 <0 .0 5 )。结论  IGFBP- 1和 IGFBP- 2可能都参与了孕早期大鼠子宫内膜蜕膜化 ,胎盘形成及胚胎生长发育的调节 ,P可通过 PR诱导 IGFBP- 1和 IGFBP- 2在大鼠子宫内膜的表达 ,但 IGFBP- 1和 IGFBP- 2究竟是作为 P的中介因

Abstract:Objective To detect the expression of insulin like growth factor binding factor 1 and 2(IGFBP 1, 2) in endometrium and their relationships with serum progesterone(P) during peri implantation in rat. Methods The model of rat during 3 10 days of pregnancy(3d 10d) was established, and the rat in estrus was used as control. The serum P level was measured by radioimmunoassay.The expression of IGFBP 1,IGFBP 2 and progesterone receptor(PR) in endometrium and trophoblast cell was detected by histoimmunochemistry and quantified by computer image analysis. Results The serum P of pregnant rat appeared like a wave and reached peak during 5d to 8d. PR mainly expressed in luminal and glandular epithelium, endometrial stromal cell and trophoblast cell,the intensity of its expression in endometrium also appeared like wave and showed significant correlation with serum P, and the intensity of PR expression in trophoblast cell increased with the days of pregnancy and showed no significant correlation with serum P. The positive cells expressing IGFBP 1 and IGFBP 2 in endometrium of rat were similar to those expressing PR, but in trophoblast the expression of IGFBP 1 and IGFBP 2 was not detected. The intensity of IGFBP 1 expression in endometrium of rat varied like that of PR and showed significant correlation with serum P. However, the intensity of IGFBP 2 expression increased and remained stable after the rat was proved pregnant, it decreased to the level of estrus during 8d to 9d and then increased quickly on the following 10d.Conclusion IGFBP 1 and IGFBP 2 both play roles in the modulation of endometrial decidulization, placenta formation, and embryo growth and development; their expression can be induced by P during early pregnancy; yet, it remains not clear whether IGFBP 1 and IGFBP 2 act as the mediator or the cofactor of progesterone.
